Pim-3 expression was elevated in human gastric cancer cell lines and was correlated with the aggressiveness of gastric carcinoma. Pim-3 was localized in the cytoplasm and was required for cell cycle progression. Pim-3 is essential for tumor cell proliferation and anchorage-independent growth, and it may contribute to the metastatic potential of cancer cells. Pim-3 inhibits the apoptosis of gastric cancer cells, and it enhances cell-extracellular matrix (ECM) and cell-cell adhesion. Mice with Pim-3 knockdown show reduced growth rates of subcutaneous and metastatic tumors. In vivo functional analysis demonstrated that Pim-3 promotes tumor growth and metastasis in the mouse tumor xenograft model. Inhibition of Pim-3 suppresses angiogenesis and up-regulates matrix metalloproteinase (MMP)-9 expression in vivo. Our study indicates that Pim-3 plays an oncogenic role in gastric cancer and may represent a potential therapeutic target for this disease
